Dreidimensionale konfokale Absorptionsmessungen zur räumlichen Strukturaufklärung
This study introduces a new measurement procedure for three-dimensional microscopy. With this procedure, image formation results merely from an induced change in transmission of certain components in a transparent sample. In order to achieve this, a mathematical model was created to explain image formation due to the photophysical backgrounds based on a simple dye system. Using this model, the attainable spatial resolution and sensitivity of detection of this measurement technique can be determined. Furthermore, the model forecasts an alternative method to examine the molcecular dynamics. Also contained is a description of a setup for the experimental verification of the individual forecasts of the model. The study describes measurements to verify the individual forecasts of the model and to indicate the prospects of the procedure. The tests demonstrate that the spatial imaging of non-fluorescent specimen with high resolution is possible. Furthermore, the study contains an estimation of the attainable detection sensitivity which can be achieved with commercialized light sources for measurement currently available. The study also shows that the dynamics of transient decay processes can be examined using this procedure. Based on these studies, the University of Siegen in Germany has successfully filed a patent in 2002.
